چکیده
Previous evaluation of HWP1 in systemic candidiasis in CBA/J mice was done with Candida albicans strains with differing genetic locations of URA3 as a result of Ura-blaster mutagenesis. In this study, the presence of HWP1 and the location of URA3 contributed to the severity of murine systemic candidiasis in BALB/c mice.

Flanking direct repeats of hisG alter URA3 marker expression at the HWP1 locus of Candida albicans.
HWP1 encodes an adhesin of Candida albicans and has been implicated in filamentation and virulence. URA3, an often-used transformation selection marker, is apparently incorrectly expressed when integrated at the HWP1 locus, which results in an attenuated virulence phenotype. Expression of URA3 is compromised by ectopic integration at other loci as well. In contrast, prior studies from the autho...
متن کاملNiche-Specific Requirement for Hyphal Wall protein 1 in Virulence of Candida albicans
Specialized Candida albicans cell surface proteins called adhesins mediate binding of the fungus to host cells. The mammalian transglutaminase (TG) substrate and adhesin, Hyphal wall protein 1 (Hwp1), is expressed on the hyphal form of C. albicans where it mediates fungal adhesion to epithelial cells. Hwp1 is also required for biofilm formation and mating thus the protein functions in both fung...
متن کاملEvaluation of the roles of four Candida albicans genes in virulence by using gene disruption strains that express URA3 from the native locus.
Reintroducing URA3 to its native locus in Candida albicans not5, not3, bur2, and kel1 disruption mutants enabled us to directly compare strains with control strain CAI-12. We showed that URA3 position affected orotidine 5'-monophosphate decarboxylase activity, hyphal morphogenesis, adherence, and mortality in murine disseminated candidiasis. After URA3 was reintroduced to its native locus, only...
متن کاملHwp1 gene Expression of Candida albicans and Study its role in adherence
Candida albicans is an opportunistic fungal pathogen found as mycoflora in the human body surfaces. Sevral genes play a crucial rule in its virulence including Hwp1 (hyphal wall protein 1), BCR1 and ALS gene family. Hwp1 gene is a responsible for coding a cell surface protein, which mediates biofilm formation in candida albicans. Here we investigated the presence of the HWP1gene was characteris...
متن کاملAltered expression of selectable marker URA3 in gene-disrupted Candida albicans strains complicates interpretation of virulence studies.
The ura-blaster technique for the disruption of Candida albicans genes has been employed in a number of studies to identify possible genes encoding virulence factors of this fungal pathogen. In this study, the URA3-encoded orotidine 5'-monophosphate (OMP) decarboxylase enzyme activities of C. albicans strains with ura-blaster-mediated genetic disruptions were measured. All strains harboring gen...
